Mechanism of Action

Pregabalin does not function through traditional GABAergic mechanisms. Instead, it selectively binds to the alpha2-delta protein on voltage-gated calcium channels within over-excited neurons. This binding reduces calcium influx at nerve terminals and inhibits the release of excitatory neurotransmitters. As a result, it effectively dampens hyperexcitable neuronal circuits associated with neuropathic pain, epilepsy, and anxiety.

Recommended dose of Pregabalin 150 mg is once or twice daily, depending on indication and patient tolerance.

For neuropathic pain, initial dosing may begin at 75 mg twice daily, escalating to 150 mg BID.

Dose adjustments may be required in patients with renal impairment, as Pregabalin is eliminated through the kidneys.
